RESUMO: Apesar da reconhecida importância de Clostridium perfringens como enteropatógeno de pequenos ruminantes, pouco se sabe sobre a frequência dos genótipos ou do papel de fatores de virulência adicionais de C. perfringens em cabras saudáveis, dificultando o diagnóstico laboratorial da infecção causada por esse micro-organismo. Dessa forma, o presente estudo teve como objetivo caracterizar C. perfringens de amostras de fezes de cabras adultas saudáveis. Amostras de fezes de 250 cabras saudáveis de 17 rebanhos diferentes em Minas Gerais, Brasil, foram submetidas ao isolamento e genotipagem de C. perfringens. C. perfringens tipo A foi isolado de 189 (75,6%) cabras, enquanto C. perfringens tipos C e D foram detectados em um animal (0.4%) cada. Todos os isolados foram negativos para os genes codificadores das toxinas NetB, NetE, NetF e enterotoxina. Os resultados apresentados confirmam C. perfringens tipo A como parte da microbiota de cabras saudáveis e sugere que C. perfringens tipos C e D são raramente encontrados em caprinos saudáveis.

ABSTRACT: Despite the known importance of Clostridium perfringens as an enteropathogen in small ruminants, little is known about the role of its additional virulence factors or the frequency of the various C. perfringens genotypes in healthy goats; this complicates the laboratory diagnosis of the infections caused by this microorganism. In light of this, the aim of the present study was to isolate and genotype C. perfringens from stool samples from healthy goats in Brazil. Stool samples from 250 apparently healthy adult goats from 17 different herds in Minas Gerais, Brazil were collected, and isolation and genotyping of C. perfringens was performed. C. perfringens type A was isolated from 189 (75.6%) goats, whereas C. perfringens types C and D were each detected in one goat (0.4%). All isolates were negative for enterotoxin-, NetB-, NetE-, and NetF-encoding genes. These results confirmed C. perfringens type A as part of the microbiota in these animals, and they suggested that C. perfringens type C and D are rarely isolated from healthy goats.

BACKGROUND The World Health Organization (WHO) has classified human zoonotic tuberculosis (TB) due to Mycobacterium bovis as a neglected issue in the developing world. In a recent cross-sectional study in Brazil, three of 189 TB patients presented with a coinfection of M. bovis and M. tuberculosis and were selected as cases for this study. OBJECTIVE The aim was to evaluate risk factors (RF) for zoonotic TB in an urban area of Brazil in order to guide preventive programmes. METHODS A matched case-control study was carried out nested within a cross-sectional study. For each of the three cases, 14 age- and sex-matched controls (TB due to M. tuberculosis) were selected. FINDINGS Zoonotic potential exposures (ZE) and extrapulmonary TB (EPTB) were independently associated with zoonotic TB in multivariate analyses. CONCLUSIONS ZE by occupation and consumption of raw milk and derivative products that place individuals in direct and indirect contact with animals and their excretions/secretions increase the risk for zoonotic TB in Brazil, especially among those with EPTB. Therefore, measures such as efficient control of bovine TB, distribution of pasteurised milk and its derivative products, and the diagnosis and monitoring of zoonotic TB in humans are essential steps, especially in developing countries where bovine TB is enzootic, and further studies are necessary.

CONTEXTO E OBJETIVO: Mycobaterium avium subsp. paratuberculosis (MAP) tem atraído o interesse de pesquisadores devido às semelhanças entre a paratuberculose e a doença de Crohn (CD). Este estudo objetivou avaliar a frequência de MAP por meio de cultura, histologia e reação da polimerase em cadeia (PCR), em biópsias intestinais de pacientes brasileiros com CD. PCR quantitativa em tempo real (qRT-PCR) foi realizada nas amostras positivas. TIPO DE ESTUDO E LOCAL: Estudo transversal analítico com grupo controle realizado em duas universidades federais. MÉTODOS: Amostras frescas foram coletadas de 25 pacientes; cinco com CD, oito com colite ulcerativa (UC) e 12 controles sem doença inflamatória intestinal (nIBD). Também foram coletadas 149 amostras fixadas em parafina (FFPE): 44 CD, 49 UC e 56 nIBD. RESULTADOS: Nenhuma das amostras frescas foi positiva para MAP. Cinco amostras FFPE (uma CD, duas UC e duas nIBD) e três amostras frescas (uma de cada grupo) foram positivas por IS900-PCR. qRT-PCR foi realizada nessas oito amostras. Nas amostras FFPE, havia 192,12 cópias/μl no grupo CD, 72,28 cópias/μl no UC e 81,43 cópias/μl no nIBD. Nas amostras frescas, havia 432,99 cópias/μl, 167,92 cópias/μl e 249,73 cópias/μl nos grupos CD, UC e nIBD, respectivamente. A maior carga bacteriana foi encontrada no grupo CD. CONCLUSÃO: Este estudo não fornece evidências do papel de MAP na etiologia da CD, embora DNA de MAP tenha sido detectado em pacientes dos três grupos. Este é o primeiro relato da presença de MAP em biópsias intestinais humanas no Brasil.

CONTEXT AND OBJECTIVE: Mycobacterium avium subsp. paratuberculosis (MAP) has attracted the interest of researchers because of similarities between paratuberculosis and Crohn's disease (CD). The aim of this study was to evaluate the frequency of MAP through cultures, histology and polymerase chain reaction (PCR) on intestinal biopsies from Brazilian CD patients. Quantitative real time PCR (qRT-PCR) was performed on positive samples. DESIGN AND SETTING: Analytical cross-sectional study with control group at two federal universities. METHODS: Fresh samples were collected from 25 patients; five with CD, eight with ulcerative colitis (UC) and 12 controls with non-inflammatory bowel disease (nIBD). Formalin-fixed paraffin-embedded (FFPE) samples from 143 patients were also collected: 44 CD, 49 UC and 56 nIBD. RESULTS: None of the fresh samples was positive for MAP. Five FFPE samples (one CD, two UC and two nIBD) and three fresh samples (one in each group) were positive through IS900-PCR. qRT-PCR was performed on these eight samples. Among the FFPE samples, there were 192.12 copies/μl in the CD group, 72.28 copies/μl in UC and 81.43 copies/μl in nIBD. Among the fresh samples, there were 432.99 copies/μl, 167.92 copies/μl and 249.73 copies/μl in the CD, UC and nIBD groups, respectively. The highest bacterial load was in the CD group. CONCLUSION: This study does not provide evidence for a role of MAP in the etiology of CD, although MAP DNA was detected in all three patient groups. This is the first report of MAP presence in human intestinal biopsies in Brazil.

O herpesvírus bovino 1 (Bovine herpesvirus 1 - BoHV-1) causa grandes prejuízos econômicos à pecuária bovina mundial, por comprometer os sistemas respiratório e reprodutivo de bovinos de corte e leiteiros. Nas últimas décadas, os registros de casos clínicos ocasionados pelo BoHV-1, no Brasil, mostram que têm aumentado gradualmente, o que torna importante o delineamento de estudos que mostrem a distribuição da infecção nas diversas regiões do país. O objetivo deste estudo foi investigar a frequência de anticorpos anti-BoHV-1 em rebanhos bovinos leiteiros do Estado do Espírito Santo (ES), região ainda carente de informações sobre a distribuição desse patógeno. Para o estudo, foram utilizadas amostras de soro de bovinos leiteiros, não vacinados contra o BoHV-1, de 1.161 animais, provenientes de 59 rebanhos, oriundos de 23 municípios pertencentes às macrorregiões metropolitana, norte, noroeste e sul do ES. Para a identificação de animais soropositivos, foi aplicada a técnica de vírus-neutralização em microplacas. Das amostras de soros avaliadas, 775 (66,75%) apresentaram anticorpos neutralizantes contra BoHV-1, sendo todos os rebanhos considerados positivos. As percentagens das frequências de animais positivos foram bastante variadas, apresentando 49,06, 62,15, 67,21 e 80,04%, para as macrorregiões metropolitana, sul, norte e noroeste, respectivamente. Neste estudo, os resultados claramente indicam a circulação do agente viral no rebanho bovino leiteiro do ES, sendo necessário o controle e monitoramento das enfermidades relacionadas com o BoHV-1.

Bovine herpesvirus 1 (BoHV-1) causes major losses in worldwide livestock, affecting the respiratory and reproductive tracts of bovine. In the past decades, the number of cases in Brazil has been gradually increasing. Therefore, it is important to assess the distribution of infection in different regions of the country. In the state of Espírito Santo (ES) the BoHV 1 infection rate in dairy cattle herds is unknown. Thus, the aim of this study was to detect neutralizing antibodies against BoHV-1 in serum samples from 1,161 non-vaccinated cows from 59 dairy cattle herds in 23 municipalities of the Metropolitan, North, Northwest and South macro-regions. The identification of seropositive cows was evaluated by the virus neutralization test. The results showed that of all serum samples evaluated 775 (66.75%) had neutralizing antibodies against BoHV-1. Moreover, all herds were found positive; however, the percentage of positive cows varied among regions; 49.06%, 62.15%, 67.21% and 80.04% for the Metropolitan, South, North and Northwest macro-regions, respectively. In this study, the results clearly indicate the dissemination of the viral agent in dairy cattle in the ES state, requiring the monitoring and control of diseases related to BoHV-1 infection.

Biofilmes bacterianos estão envolvidos no agravamento e recorrência de mastite clínica em rebanhos leiteiros. Vários fatores, tais como o pH, temperatura, concentração de O2 e glicose podem afetar sua indução e suas taxas de crescimento. Neste estudo, a produção de biofilme foi demonstrada por 27 isolados de Escherichia coli de mastite bovina a valores de pH diferentes, dependendo da disponibilidade de glucose, simulando as condições encontradas nas glândulas mamárias afetadas pela doença. A formação de biofilme foi analisada por espectrofotometria em microplaca com 16 diferentes meios de cultura e por microscopia eletrônica de varredura. A formação de biofilmes foi maior em isolados cultivados em condições associadas com baixa disponibilidade de glucose (0,5% ou 1,5%) tanto com pH ácido (5,5) como alcalino (8,5), em comparação com as condições associadas com a disponibilidade de alto teor de glucose (2,5% ou 3,5% ) e pH próximo à neutralidade (6,5 ou 7,5). Os resultados indicam o possível favorecimento da produção de biofilmes em fases posteriores do processo infeccioso de mastite causada por E. coli, quando o ambiente da glândula é menos propício ao crescimento bacteriano devido às condições de estresse acima mencionados, em contraste com o ambiente da glândula mamária saudável, no qual não há nenhuma limitação de nutrientes ou condições de alcalinidade ou acidez em particular. Ainda, o conhecimento da fase em que se encontra a infecção e as condições ambientais da glândula que causariam maior produção de biofilmes é de grande importância para orientar as estratégias de controle mais adequadas para prevenir recidivas após o tratamento da mastite bovina, uma doença economicamente importante na bovinocultura leiteira em todo o mundo.

Bacterial biofilms are involved in the aggravation and recurrence of clinical mastitis in dairy herds. Several factors such as pH, temperature, concentration of O2 and glucose can affect their induction and growth rates. In this study, biofilm production was demonstrated by 27 Escherichia coli strains isolated from bovine mastitis at different pH values depending on the availability of glucose, mimicking conditions found in mammary glands affected by the disease. Biofilm formation was analyzed by spectrophotometric analysis in microtiter plate with 16 different culture media and by scanning electron microscopy. Biofilm formation was greater in isolates cultured under conditions associated with low glucose availability (0.5% or 1.5%) and with either an acidic (5.5) or alkaline (8.5) pH, compared to conditions associated with high glucose availability (2.5% or 3.5%) and near-neutral pH (6.5 or 7.5). Results indicate possible favoring of biofilm production in the later stages of the infectious process caused by E. coli, when the gland environment is less propitious to bacterial growth due to the stress conditions mentioned above; contrasting with the environment of the healthy mammary gland, in which there is no limitation on nutrients or conditions of particular alkalinity or acidity. Thus, knowledge of the stage in which is the infection and environmental conditions of the mammary gland that cause increased production of biofilms is of paramount importance to guide the most appropriate control strategies to prevent relapse after treatment of bovine mastitis, an economically important disease in dairy cattle worldwide.

Este estudo avaliou a atividade inibitória do óleo de copaíba (Copaifera officinalis contra o microrganismo cariogênico, Streptococcus mutans. Para isso, foi realizado um teste de concentração mínima inibitória do óleo de copaíba contra S. mutans, utilizando a técnica de diluição seriada em caldo, com um controle negativo, um controle positivo (clorexidina a 0,12%) e uma solução de óleo de copaíba 10% como teste. Também foi conduzido um teste de concentração mínima bactericida com os tubos que apresentaram inibição microbiana. No teste de concentração inibitória mínima, o óleo de copaíba mostrou inibição do crescimento bacteriano em todas as concentrações testadas até 0,78 µL/mL da solução a 10% do óleo de copaíba no caldo. Além disso, o controle negativo não teve nenhuma inibição, e a solução de clorexidina 0,12% foi eficaz até 6,25 µL/mL no caldo. O óleo de copaíba mostrou uma atividade bacteriostática contra S. mutans em baixas concentrações, apresentando-se assim como uma opção de fitoterápico a ser utilizado contra bactérias cariogênicas na prevenção de cáries.

This study evaluated the inhibitory activity of copaiba oil (Copaifera officinalis against the cariogenic microorganism, Streptococcus mutans. For such purpose, a minimum inhibition concentration test of copaiba oil against S. mutans was performed, using the serial dilution in broth technique, with a negative control, a positive control (0.12% chlorhexidine) and a 10% copaíba oil solution as a test. A minimum bactericidal concentration test with tubes presenting microbial inhibition was also conduced. In the minimum inhibitory concentration test, copaiba oil showed inhibition of bacterial growth at all concentrations tested up to 0.78 µL/mL of the 10% copaiba oil solution in the broth. In addition, the negative control had no inhibition, and the 0.12% chlorhexidine solution was effective up to 6.25 µL/mL in the broth. Copaiba oil showed a bacteriostatic activity against S. mutans at low concentrations, and could be a an option of phytotherapic agent to be used against cariogenic bacteria in the prevention of caries disease.

Os objetivos deste estudo foram avaliar o efeito antimicrobiano de diferentes concentrações de óleo de copaíba contra o crescimento de Listeria monocytogenes e analisar as diferenças na inibição do microorganismo com soluções do óleo autoclavadas e não autoclavadas. Para tanto, foi realizado um teste de difusão em ágar, com seis isolados do microrganismo originários de produtos cárneos, seis concentrações de ambas as soluções e um controle negativo. Os resultados mostraram sensibilidade de cinco cepas de L. monocytogenes em relação à solução 10% de óleo de copaíba autoclavada. Quatro isolados também apresentaram sensibilidade para a solução de 5% autoclavada e apenas um foi sensível à solução de 2,5% autoclavada. A solução 10% não autoclavada apresentou inibição do crescimento de apenas dois isolados. Esses resultados apontaram a solução autoclavada de 10% do óleo de copaíba com maior inibição em relação a todas as outras soluções e concentrações testadas (P<0,05). As soluções 5 e 2,5% autoclavadas e a solução 10% não autoclavada apresentaram-se estatisticamente iguais. Todas as outras concentrações das soluções de copaíba e o controle negativo não apresentaram inibição das bactérias. Os resultados deste estudo sugerem que o óleo de copaíba autoclavado pode ser uma potencial fonte de novos agentes para o controle de infecção ou para a conservação dos alimentos, inibindo o crescimento de bactérias de origem alimentar como L. monocytogenes.

The aim of this study was to evaluate the antimicrobial effect of different copaiba oil concentrations against the growth of Listeria monocytogenes, and analyze differences in inhibition of microorganisms with autoclaved and non autoclaved oil. This study provided an agar diffusion test with six isolates of bacteria and six different concentrations of autoclaved or non autoclaved copaiba oil and a negative control. The results showed sensitivity of five L. monocytogenes isolates related to the 10% autoclaved solution of copaiba oil. Four strains also showed sensitivity to the 5% autoclaved solution and one to 2.5% autoclaved solution. The 10% non autoclaved oil solution showed growth inhibition only for two strains. These results had pointed the 10% autoclaved solution of copaiba oil with higher inhibition as all other solutions and concentrations tested (P<0.05). For the other concentrations of both solutions, the 5 and 2.5% autoclaved and 10% non autoclaved solutions had presented statistically equal. All other concentrations of both copaiba solutions and the negative control did not presented any bacteria inhibition. In conclusion, the results of this study suggest that the autoclaved copaiba oil may be a potential new agent source for infection control or for food preservation, inhibiting the growth of food-borne bacteria such as L. monocytogenes.

Os membros da família Enterobacteriaceae estão presentes no intestino do homem e dos animais como comensais ou agentes causadores de doença importantes. Bactérias multirresistentes podem possuir sistemas de efluxo multidrogas (MDR) sendo capazes de sobreviver em nichos ecológicos adversos. Escherichia coli e Enterobacter cloacae, multirresistentes, isoladas de frangos sadios foram investigadas quanto à presença de MDR. A diminuição da concentração inibitória mínima de antimicrobianos, na presença de um desacoplador da força próton motora (PMF), foi usada para detectar o fenótipo MDR. Foi realizada PCR dos genes codificadores de AcrA e AcrB, componentes de um sistema MDR. A diversidade de cada isolado foi confirmada por eletroforese em gel de campo pulsado (PFGE) usando a endonuclease XbaI. Observou-se em todos os isolados, exceto E. coli 1 e E. cloacae 9, uma diminuição das MICs ou das curvas de crescimento na presença dos antimicrobianos, indicando um mecanismo de resistência dependente da PMF. Os produtos amplificados esperados derivados de acrAB foram obtidos em todos os isolados de E. coli e em dois, dos cinco, de E. cloacae. O dendrograma gerado mostra diferentes perfis de bandas (pulsetypes), confirmando a diversidade genética entre os isolados. Uma questão importante e relacionada à saúde publica é o fato de que diferentes modelos e mecanismos de resistência aos antimicrobianos estão presentes em um número reduzidos de isolados não patogênicos e obtidos de uma mesma origem. Esses podem ser fontes de genes de resistência para outros microorganismos, entre eles, cepas patogênicas.

Members of the Enterobacteriaceae family are present in the intestines of man and animals as commensals or are important disease causing agents. Bacteria bearing multidrug efflux systems (MDR) are able to survive adverse ecological niches. Multiresistant Escherichia coli and Enterobacter cloacae isolates from wholesome broiler carcasses were investigated for the presence of MDR. Lowering of Minimal Inhibitory Concentration for antimicrobials in the presence of a proton-motive force (PMF) uncoupler was tested as a potential display of the MDR phenotype. PCR amplification of the genes encoding AcrA and AcrB, components of a MDR system was performed. Diversity of each species was ascertained by Pulsed-Field Gel Electrophoresis (PFGE) of DNA digested with endonuclease XbaI. For all the isolates, except E. coli 1 and E. cloacae 9, lowering of MIC or of the growth rate in the presence of antimicrobials was observed, indicating a PMF dependent resistance mechanism. Expected products of DNA amplification with acrAB derived primers was obtained with all E. coli strains and with two of the five E. cloacae strains. Dendrogram generated shows diverse pulsetypes, confirming the genetic diversity among the strains. An important issue and related public health is the fact that different models and mechanisms of antimicrobial resistance are present in a small number of non-pathogenic strains and isolated from the same origin. These may be sources of resistance genes to others microorganisms, among them, pathogenic strains.

Pênfigo foliáceo é uma rara afecção cutânea auto-imune que acomete várias espécies. Relata-se o caso de uma égua de quatro anos de idade, sem raça definida, apresentando áreas de alopecia, exsudação e crostas localizadas na face, região peitoral e membros pélvicos, além de prurido. Os exames laboratoriais para pesquisa de ácaros, bactérias e estruturas fúngicas foram negativos. O exame histopatológico revelou uma dermatite pustular intra-epidérmica com acantólise subcorneal, assim como dermatite perivascular superficial, com infiltração de eosinófilos, sendo o quadro compatível com pênfigo foliáceo.

Pemphigus foliaceus is an uncommon autoimmune skin disorder affecting various species. A case of a 4-year-old mare of undefined breed, displaying alopecic areas, exsudation and crusty lesions on the face, breast region, and hindlimbs as well as pruritus, is reported. Laboratory exams for mite, bacteria and fungal structures were negative. Histopathological exam revealed an intra-epidermal pustule dermatitis with acantholysis, as well as superficial perisvascular dermatitis with infiltration of eosinophyles, conditions compatible with pemphigus foliaceus.

Objetivou-se estudar a viabilidade da substituição parcial ou total do nitrato de amônio por uréia como fonte de nitrogênio no meio de cultura para o cultivo in vitro do abacaxizeiro cv. Pérola. Plantas oriundas das gemas da coroa do fruto com massa fresca em torno de 80 mg foram inoculadas em meio MS (líquido e sólido) substituindo-se 100, 80, 60, 40, 20 e 0% do nitrato de amônio por uréia. O delineamento experimental utilizado foi o inteiramente casualizado, com quatro repetições e três plantas/repetição. Após 60 dias, observou-se a possibilidade da substituição parcial de 40% de nitrato de amônio por uréia para as variáveis analisadas e melhor desenvolvimento das plantas em meio de cultura sólido.

The partial or total substitution viability of the ammonium nitrate for urea, as source of nitrogen in the culture medium for the in vitro culture of pineapple cv. Pérola was studied. Plants originating from fruit crown buds with fresh weight matter around 80 mg was inoculated on MS medium (liquid and solid) with substitution of 100, 80, 60, 40, 20 and 0% of the ammonium nitrate for urea. These treatments were arranged in a completely randomized design, with four replications and three plants/replications. After 60 days, the possibility of 40% ammonium nitrate for urea partial substitution was observed for the analyzed variables. In addition, better plant development in solid culture medium was observed as well.

Objetivou-se com este trabalho verificar o efeito da matéria orgânica como componente de substratos para mudas micropropagadas de abacaxizeiro cv. Pérola em fase de aclimatização. Mudas micropropagadas de abacaxizeiro cv. Pérola, selecionadas de acordo com o peso (aproximadamente 2 g), foram plantadas em bandejas de isopor de 72 células de 120 cm³ contendo proporções de substratos com terra, esterco bovino, Plantmax e matéria orgânica. O delineamento estatístico utilizado foi o inteiramente casualizado com 4 repetições e 3 plantas por parcela. As avaliações foram feitas 90 dias após o plantio, quando se avaliou: altura da planta, comprimento de raiz, peso de matéria fresca de raiz e parte aérea, peso de matéria seca de raiz e parte aérea e número de folhas. Com os resultados obtidos, conclui-se que a matéria orgânica tem efeito significativo no desenvolvimento das mudas. Com o substrato contendo terra, esterco e Plantmax foi obtido o melhor desenvolvimento das raízes e parte aérea.

The objective of this work was to evaluate the effect of the organic matter as component of substrata for micropropagated plants of pineapple cv. Pérola in the acclimatization phase. Micropropagated plants were selected according to the weight (approximately 2g) and planted in trays with 72 cels of 120 cm³, containing substrata made of soil, bovine manure, Plantmax and organic matter at different proportions. At 90 days after planting, the following parameters were evaluated: height of the plant, root length, weight of fresh matter of root and aerial part, dry matter weight of root and aerial part and number of leaves. The obtained results allowed to conclude that the organic matter has significant effect on the development of the seedlings. The best results for the development of the roots and aerial part were obtained with substratum containing soil, manure and Plantmax.

Multidrug efflux mechanisms in bacteria contribute significantly to intrinsic and acquired resistance to antimicrobial agents. Genome analysis have confirmed the broad distribution of these systems in Gram-negative as well as in Gram-positive bacteria. Among resistance mechanisms, the multidrug efflux system or pump deserves special attention, since a cell that has acquired it can simultaneously diminish or even suppress the susceptibility to a wide range of antimicrobials. The efflux system is mediated by transport proteins which confer resistance to toxic compounds. In Gram-negative bacteria, a tripartite efflux system is necessary to expel the drug to the outer medium: a protein localized in the cytoplasmic membrane; another in the periplasmatic space (membrane fusion protein - MFP); and a third in the outer membrane (outer membrane factor - OMF). The drug transport is active, and depends either on the energy provided by ATP hydrolysis or is directly driven by the proton motive force. The transport proteins are grouped in families, according to the homology of the amino acid sequences and to similarity of mechanisms. Among Gram-negative bacteria, Escherichia coli and Pseudomonas aeruginosa have most of the hitherto identified and studied multidrug efflux systems.

Objetivou-se estudar o efeito do estiolamento na micropropagação de abacaxizeiro cv. Pérola e posterior recuperação dos brotos estiolados, realizando-se dois experimentos. No primeiro, os caules utilizados como explantes foram obtidos de brotos pré-estabelecidos in vitro, dos quais foram retiradas as folhas. O estiolamento foi induzido colocando-se os explantes em tubos de ensaio no escuro por 20, 40 e 80 dias, contendo os seguintes meios de cultura: 1) MS suplementado com 0,1 mg.L-1 de ANA e 0,5 mg.L-1 de BAP, 2) MS suplementado com 1,8 mg.L-1 de ANA e 2 mg.L-1 de BAP e 3) MS sem reguladores de crescimento. Para número de brotos, o melhor meio foi o MS + 1,8 mg.L-1 de ANA e 2 mg.L-1 de BAP, obtendo-se média de 10,26 brotações aos 40 dias no escuro. Para comprimento de brotos estiolados, o meio MS sem reguladores de crescimento foi significativamente melhor quando os explantes foram mantidos por 80 dias no escuro, apresentando comprimento médio de 10,86 cm. No segundo experimento, os brotos estiolados, com e sem o ápice, foram colocados horizontalmente em placas contendo meios de cultura com idêntica formulação aos descritos anteriormente. Para número total de brotos, brotações estioladas sem ápice em meio MS suplementado com 1,8mg.L-1 de ANA e 2 mg.L-1 de BAP promoveram o melhor resultado, correspondendo a 10,61 brotações por explante.

It was aimed to produce micropropagated plantlets of pineapple cv. Pérola by using the etiolated technique and subsequent recovery of etiolated shoots. Two experiments were carried out. In the first, the stalks used as explant were obtained from in vitro shoots established without leaves. The etiolation was induced by putting the explants in test tubes in the darkness for 20, 40 and 80 days with the media: 1) MS supplemented with ANA 0.1mg.L-1 and BAP 0.5mg.L-1, 2) MS supplemented with ANA 1.8mg.L-1 and BAP 2mg.L-1 and 3) MS without growth regulators. The best results for shoot number were obtained with MS + ANA 1.8mg.L-1 and BAP 2mg.L-1 media (10.26 shoots at 40 days in the darkness). The MS medium without growth regulators, was significantly better for etiolated shoots when the explants were maintained for 80 days in the darkness, presenting 10.86cm mean length. In the second experiment, the etiolated shoots, with or without apex, were put horizontally in plates containing the three culture media described before. The MS medium supplemented with ANA 1.8mg.L-1 and BAP 2mg.L-1 induced the best result for total shoots number (10.61 shoots when the apex were eliminated).

Foi realizado um estudo clínico-epidemiológico da esquistossomose em escolares (6 - 14 anos) da Ilha, em Minas Gerais (Brasil). Foram feitos exame parasitológico de fezes pelo método de KATO-KATZ e exame clínico em, respectivamente, 86,7 e 85,4% da população escolar. Foi realizado levantamento sócio-econômico e foram pesquisados os sinais e sintomas apresentados pelos pacientes e seus contatos com águas naturais. O índice de infecção pelo Schistosoma mansoni foi 32,7%, predominaram as baixas contagens de ovos nas fezes (89,0% dos pacientes eliminavam menos de 500 ovos/grama de fezes) e o índide de esplenomegalia foi 7,7%. Os pacientes positivos (com ovos de S. mansoni nas fezes) foram comparados aos negativos (sem ovos nas fezes e com intradermoreação negativa), verificando-se que a infecção pelo S. mansoni era significativamente mais freqüente entre os trabalhadores rurais, entre os que residiam em casas de pior qualidade e entre aqueles cujos chefes de família eram analfabetos. Constituíram os maiores fatores de risco para a infecção pelo S. mansoni nesta área: a) ter contatos com águas naturais para trabalhar na lavoura (Odds Ratio = 18,08); b) ter contatos diários com águas naturais (OR = 13,82) e c) ter contatos com águas naturais para pescar, nadar e/ou brincar (OR = 7,75 e 5,51, respectivamente). Os autores levantam a hipótese de que a transmissão da esquistossomose nesta localidade não ocorre no peridomicílio, mas sim nas lagoas próximas à Ilha e nas plantações agrícolas, provavelmente nas culturas de arroz de várzea.

A schistosomiasis mansoni clinical-epidemiological study was carried out among school children (6-14 years old) of the Ilha community, Minas Gerais. Stool examination, using the KATO-KATZ method, and clinical examination were performed on 86.7 and 85.4%, respectively, of the population studied. The signs and symptoms presented by the patients as well as their contacts with untreated water were investigated. A socio-economic survey was carried out in the community through household interviews. The index of infection by the Schistosoma mansoni was 32.7%, low egg counts were predominant (89.0% of the patients eliminated less than 500 eggs/gram of feces) and the index of splenomegaly was 7.7%. The positive patients (presenting S. mansoni eggs in the stools) were compared with the negative ones .(without eggs in the stools and presenting a negative intradermal reaction). The S. mansoni infection was significantly higher among the rural workers, among those who lived in poor housing conditions and among those patients the head of whose family was illiterate. Increased risk factors were found to be: a) the contact with untreated water used for agriculture (OR = 18.08); b) the occurrence of daily contact with untreated water (OR = 13.82) and c) the contact with untreated water for fishing, swimming and/or playing (OR = 7.75 and 5.51 respectively). The authors consider the hypothesis that the schistosomiasis transmission in this area does not occur in household surroundings but in the nearby lagoons and agricultural areas instead (probably in irrigated rice plantations).
